Immunological characteristics of the transgenic mice expressing a cytokine in the skin for the production of skin grafted animals

Abstract
IL-10 inhibits macrophage-dependent antigen presentation, a variety of cytokine production, and generation of allospecific cells in vitro. These findings have lead to the widespread expectation that IL-10 may be a useful immunosupressive agent to inhibit allograft rejection or autoimmunity in vivo.
In the present study, to understand the immunological characteristics of the transgenic mice expressing a IL-10 in the skin, we have generated transgenic mice carrying mouse IL-10 gene under control of the human keratin 14 promotor. Most founder mice died between 2 to 3 weeks of age. Death may be associated with the excess of endogenous IL-10. To study the effect of the expression of cytokines mRNA in the skin of transgenic mice, a method of semiquantitation RT-PCR was performed. The transgenic mice showed the increased expression of INF-¥ã, IL-1¥á, IL-1¥â and TNF-¥á. The results indicate that IL-10 transgenic mice might be involved in the immune system via the cytokine network.
